Mechanism of Biological Nitrogen Fixation X. Hydrogenase in Cell-free Extracts and Intact Cells of Azotobacter*

چکیده
The discovery of hydrogenase in Azotobacter cells (1) raises the question of its function in the metabolism of this organism. This enzyme is usually found in anaerobic or facultative anaerobic bacteria which liberate Hz from certain substrates. Azotobacter is a strict aerobe, and its metabolism consists primarily in the simple oxidation of substrate to carbon dioxide and water. Although not conclusive, certain observations to be discussed later suggest that hydrogenase may be related to the nitrogen fixation enzyme system in Axotobacter. Demonstration of such a relationship would provide new insight into many aspects of the mechanism of the reaction including the chemical pathway. Our previous investigations on this question have been carried out entirely with resting suspensions of the organism, but for certain studies a cell-free preparation would be advantageous. The preparation and properties of a cell-free extract from Axotobacter containing hydrogenase are described in this report.

Oxidative activity and nitrogen fixation in cell-free preparations from Azotobacter vinelandii.
Ammonia is generally conceded to be the "key intermediate" in biological nitrogen fixation-that is, the compound which reacts to form the first stable organic compound, glutamic acid (Wilson and Burris, 1953). However, there is virtually no information concerning the reactions functioning in the conversion of N2 to ammonia. Reproducible nitrogen fixation by a cell-free preparation from bacteria...

The inhibitors usually associated with the activity of the cytochrome oxidase system-cyanide and carbon monoxide-are also effective in reducing the oxidation of H(2) by intact cells of Azotobacter vinelandii. The hydrogenase system is more sensitive to CO than is the respiratory system. Oxidation of a carbon source and of hydrogen by Azotobacter cells is inhibited in a quantitatively different ...
متن کاملMechanism of Biological Nitrogen Fixation
Recently Phelps and Wilson (1) reported the occurrence of hydrogenase, the enzyme which activates molecular hydrogen, in nitrogen-fixing bacteria. Because HZ specifically inhibits nitrogen fixation by red clover plants inoculated with Rhizobium trijolii as well as by Azotobacter (2,3), possession of hydrogenase by these organisms may be more than merely fortuitous. The possibility must be consi...

Two outstanding characteristics of azotobacter, their abilitv to fix nitrogen and unusually high rates of oxidation, have stimulated interest in obtaining cell free extracts suitable for biochemical analysis of these processes. Many difficulties have been encountered in obtaining such extracts. Attempts to obtain cell free extracts capable of nitrogen fixation have given extremely inconsistent ...

The catalysis of nitrogen reduction by extracts of nitrogen-fixing organisms requires an ATPgenerating system and a source of electrons (1, 2, 5, 11, 13). The phosplhoroclastic reaction provides botlh ATP anid electrons for the reduction of nitrogen by cell-free extracts of Clostridium1t pasteurianumn (2, 5, 13) and ferredoxin functions as the natural electron carrier from the phlosphoroclastic...
